AbFluor™ 488 Annexin V Apoptosis Detection Kit from Abbkine

Annexin V (or Annexin A5) is a member of the annexin family of intracellular proteins that binds to phosphatidylserine (PS) in a calcium-dependent manner. PS is normally only found on the intracellular leaflet of the plasma membrane in healthy cells, but during early apoptosis, membrane asymmetry is lost and PS translocates to the external leaflet.

Fluorochrome-labeled Annexin V can then be used to specifically target and identify apoptotic cells, which can be detected by fluorescence microscopy or flow cytometry.

Propidium iodide (PI) is a fluorescent nucleus dye, impermeant to live cells and early apoptotic cells, but stains late apoptotic, necrotic or dead cells with red fluorescence, binding tightly to the nucleic acids in the cell.

Once binding with DNA, the excitation and emission light of PI-DNA complex is 535 nm and 615 nm.

Abbkine AbFluor™ 488 Annexin V Apoptosis Detection Kit contains Annexin V labeled with Abbkine proprietary green fluorescent dye AbFluor™ 488, which allows the identification and quantitation of apoptotic cells by flow cytometry or fluorescence microscopy. Simultaneous staining of cells with AbFluor™ 488 and propidium iodide (PI) allows the discrimination of intact cells, early apoptotic and late apoptotic or necrotic cells.

FAQ

1. Can Abbkine Annexin V Apoptosis Detection Kit detect apoptosis in animals other than humans?

Sure. Because annexin v is compatible with phosphatidylserine ( PS ), but PS has no difference among different species.

2. What’s the effect of pancreatin on adherent cells?

Pancreatin has some damage to the cell membrane. But when digested with low concentration pancreatin, the adherent cells are gently blown 2 to 3 times, centrifuged at 4 , 300 g, and 5 min, the damage caused by pancreatin can be controlled within 5 %. And there is no obvious influence on the experimental results when there is a control group.

3. Why can only use pancreatin without EDTA to digest adherent cells?

Annexin V is a Ca+ dependent protein, so EDTA cannot be added to prevent EDTA from sequestering Ca+, thus affecting annexin V and the results.

Abbkine new arrival-Annexin V Apoptosis Detection kit

Apoptosis is a form of programmed cell death to remove unwanted, damaged, or senescent cells from tissues. In normal cells, the negative phospholipids reside on the inner side of the cellular membrane while the outer surface of the membrane is occupied by uncharged phospholipids (PS). After a cell has entered apoptosis, the negatively charged PS are transported from the inner to the outer leaflet of the plasma membrane, thus exposing PS to the external cellular environment. The human anticoagulant, Annexin V, is a 35–36 kDa Ca²⁺-dependent phospholipid-binding protein that has a high affinity for PS. Annexin V labeled with a fluorophore or biotin can identify apoptotic cells by binding to PS exposed on the outer leaflet.

Propidium iodide (PI) is a membrane-impermeant DNA-binding dye that is commonly used to selectively stain dead cells in a cell population. PI is excluded by live cells and early apoptotic cells, but stains necrotic and late apoptotic cells with compromised membrane integrity. PI can be excited by the 488, 532, or 546 nm laser lines, and emits red ﬂuorescence.

CliniSciences’s First Visit to Abbkine！

On March 24, 2018, Wilfried Gay, sales manager of CliniSciences visited Abbkine to discuss future cooperation. Abbkine manager expressed a warm welcome to Wilfried and showed him around our company and customer service laboratory.

After that, Abbkine manager held a meeting to show more about our company through PPT, in which presents our development history, organizational structure and future direction, and also includes our new product release plan and marketing plan. During this, they had deep communications. Wilfried said: "it is a pleasure to work with us and hope we give more support about new products training." He also pointed out that their salesmen and customers prefer face-to-face discussion, which will deepen the promotion of brand effect. We will strengthen communications with CliniSciences to provide the necessary support timely.

We learned that Wilfried was the first time to China, and he was very impressed with Chinese cities. We invited him to have dinner for characteristic Chinese dishes. We talked about the traditional culture of China and France, having a pleasant time.

On March 25, 2018, Dan Souw, the vice president of American Research Products visited Abbkine to discuss future cooperation. On behalf of Abbkine all staff, our general manager expressed a warm welcome to Dan and showed him around our company and customer service laboratory.

During the meeting, manager Ye presented our company's development history, organizational structure and future direction in the form of PPT. Both parties had in-depth exchange of views. Dan said: "we hope that the future cooperation between the two sides will be closer, and the future cooperation plan will be put forward."

After the meeting, we invited Dan to have lunch together, and we had a good time.

CCL2 gene is one of several cytokine genes clustered on chromosome 11. Chemokines are a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of N-terminal cysteine residues of the mature peptide. This chemokine is a member of the CC subfamily which is characterized by two adjacent cysteine residues. This cytokine displays chemotactic activity for monocytes and memory T cells but not for neutrophils.
